چکیده

We read with the great interest the recent publication on a second trigeminal CGRP receptor by Walker et al. The authors convincingly present the presence of a functional noncanonical CGRP receptor (AMY1) at neuronal sites in the trigeminal system. The presence of a second CGRP receptor has been debated and although it is not yet recognized by IUPHAR, several reports have previously suggested the presence of more than one CGRP receptor. We have previously published reports on the effect of olcegepant and telcagepant in the human coronary artery. In these studies, we evaluated several concentra-

A second trigeminal CGRP receptor: function and expression of the AMY1 receptor

OBJECTIVE The trigeminovascular system plays a central role in migraine, a condition in need of new treatments. The neuropeptide, calcitonin gene-related peptide (CGRP), is proposed as causative in migraine and is the subject of intensive drug discovery efforts. This study explores the expression and functionality of two CGRP receptor candidates in the sensory trigeminal system. METHODS Recep...

Reply to comment on: A second trigeminal CGRP receptor: function and expression of the AMY1 receptor.

In their comment, Haanes et al., suggest that CGRP could act through two distinct receptors in human coronary arteries; namely the CGRP receptor (CLR/RAMP1) and the AMY1 receptor (CTR/RAMP1). 1 Although there are other possible explanations, the presence of multiple CGRP receptor subtypes could explain their elegant data. In light of the active clinical development of blocking antibodies and sm...

The effect of hypothyroidism on the trigeminal calcitonin gene-related peptide containing motoneurons: an immunohisto-chemical study in late neonatal life

CGRP, which coexists with acetylcholine in trigeminal motoneurons, could be one of the regulatory anterograde factors responsible for the enhanced expression of AChR subunits in neuromuscular junction. It has been also shown that low level of thyroid hormone plasma concentration has influence on the transitional development of muscle fibers and related motoneurons through a sever reduction in A...

Role of μ-opioid receptor in parafascicular nucleus of thalamus on morphine-induced antinociception in a rat model of acute trigeminal pain

The parafascicular nucleus (PFN) of thalamus, as a supraspinal structure, has an important role in processing of nociceptive information. In addition, μ-opioid receptor contributes to supraspinal modulation of nociception. In the present study, the effects of microinjection of naloxone (a non-specific opioid-receptor antagonist) and naloxonazine (a specific μ-opioid receptor antagonist) were in...
